General Description

  • N-Acetyl-D-mannosamine (ManNAc) is a precursor used to synthesize N-acetylneuraminic acid, the most common physiologic sialic acid present in humans and other mammals
  • Sialic acids are terminal sugars on glycoconjugates that can modify biochemistry of glycoproteins and lipids, mediate cellular adhesion, and act as immune regulators
  • Defects in sialic acid biosynthesis are associated with a number of pathologies
  • Sialic acids are often found at increased expression levels on tumor cell surfaces

Applications

  • As a sialic acid precursor, N-Acetyl-D-mannosamine can be used in many glycoengineering applications, such as modifying immune cell activity, imaging glycans, or blocking pathogen binding
  • N-Acetyl-D-mannosamine hasbeen used in preclinical laboratory studies as a treatment for hereditary inclusion body myopathy, a progressive neuromuscular disease via increased sialylation of podocalyxin
  • N-Acetyl-D-mannosamine has been used in laboratory studies to induce IgG sialylation and prevent obesity-induced hypertension in mice following high-fat diet
